Estradiol is the generic name of Vagifem.

Estradiol is a form of estrogen, a female sex hormone that regulates many processes in the body. 

Estradiol Vaginal tablets are used to relieve menopausal symptoms in the vagina such as dryness or irritation. In medical terms this is known as ‘vaginal atrophy’. It is caused by a drop in the levels of estrogen in your body. This happens naturally after the menopause.

 Estradiol Vaginal tablets (Vagifem), belongs to a group of medicines called Hormone Replacement Therapy (HRT). It works by supplementing the falling levels of estrogen in the body and thereby helps to reduce the distressing symptoms of the menopause.

How long does Estradiol Vaginal Tablets (Vagifem) take to work?

Estradiol Vaginal tablets treat the loss of vaginal estrogen, which is the underlying cause of your symptoms. Even though you may not see results right away, it begins working to help replenish vaginal estrogen when you start using it. Most bothersome vaginal symptoms are reduced between 8 - 12 weeks of treatment.

Who should not use Estradiol Vaginal Tablets (Vagifem)?

You should not use estradiol if you are allergic to it, or if you have:

  • unusual vaginal bleeding that has not been checked by a doctor;

  • liver disease;

  • a history of heart attack, stroke, or blood clot in the vein or a blood clot disorder;

  • an increased risk of having blood clots due to a heart problem or a hereditary blood disorder;

  • a history of hormone-related cancer, or cancer of the breast, uterus/cervix, or vagina; or

  • untreated endometrial hyperplasia

Estradiol Vaginal tablet is for use in postmenopausal women only. If you become pregnant, stop using this medication and contact your doctor.

What to avoid while using Estradiol Vaginal Tablets (Vagifem)?

Smoking can greatly increase your risk of blood clots, stroke, or heart attack while using estradiol. This should be avoided.

Use of other vaginal products should be avoided without your doctor's advice.

Grapefruit and its products may interact with estradiol and lead to unwanted side effects.  Avoid Grapefruit.

What should I tell my healthcare provider before using Estradiol Vaginal Tablets (Vagifem)?

Tell your doctor if you have ever had or may be:

  • heart disease;

  • liver problems, or jaundice caused by pregnancy or taking hormones;

  • kidney disease;

  • gall bladder disease;

  • asthma;

  • epilepsy or other seizure disorder;

  • migraines;

  • lupus;

  • endometriosis or uterine fibroid tumors;

  • hereditary angioedema;

  • porphyria (a genetic enzyme disorder that causes symptoms affecting the skin or nervous system);

  • a thyroid disorder; 

  • high or low levels of calcium in your blood; or

  • pregnant or breastfeeding

What other medicines interact with Estradiol Vaginal Tablets (Vagifem)?

Please tell your doctor or pharmacist if you are using or have recently used any other medicines, including medicines obtained without a prescription, herbal medicines or other natural products.

However, Estradiol Vaginal Tablets (Vagifem) is used for a local treatment in the vagina and is not likely to affect other medicines. 

Estradiol Vaginal Tablets (Vagifem) may affect other vaginally applied treatments.

What are the possible side effects of Estradiol Vaginal Tablets (Vagifem)?

Get emergency medical help if you have signs of an allergic reaction to Estradiol Vaginal Tablets (Vagifem): hives; difficulty breathing; swelling of your face, lips, tongue, or throat. Call your healthcare provider right away if you get any of the following warning signs or any other unusual symptoms that concern you: New breast lumps Unusual vaginal bleeding Changes in vision or speech Sudden new severe headaches Severe pains in your chest or legs with or without shortness of breath, weakness and fatigue

Common side effects include: Headache Breast pain Irregular vaginal bleeding or spotting Stomach or abdominal cramps, bloating Nausea and vomiting Hair loss Fluid retention Vaginal yeast infection

What happens if I overdose?


If you have used more Estradiol Vaginal tablets (Vagifem) than you should, talk to a doctor or pharmacist. The dose of estradiol is so low that a considerable number of tablets would have to be taken to approach the dose normally used for treatment taken by mouth.

This medicine may be harmful if swallowed. If swallowed or an overdose is suspected, seek emergency medical attention.
